Dgat enzyme activity
WebJan 19, 2024 · In the MG pathway, the first enzymatic step is the acylation of MG with acyl-CoA to diglyceride (DG) catalyzed by MG acyltransferase (MGAT) [ 1, 2 ]. DG is then further acylated to TG by DG acyltransferase (DGAT) [ 1, 2 ]. WebThis gene encodes a multipass transmembrane protein that functions as a key metabolic enzyme. The encoded protein catalyzes the conversion of diacylglycerol and fatty acyl CoA to triacylglycerol. This enzyme can also transfer acyl CoA to retinol. Activity of this protein may be associated with obesity and other metabolic diseases.

Webenzymes for which the activity is difficult to measure. For instance, DGAT activity is routinely determined by the acyla-tion of exogenous DAG with radiolabeled fatty acyl-CoA and using thin-layer chromatography (TLC) to separate the radiola-beled TG formed in the reaction.17,18,21 To adapt the LC/MS-based assay for DGAT1 HTS, several WebDGAT activities were measured with two different acyl acceptors and assay methods using two different acyl-CoAs, and in all cases the ratio of PDAT to DGAT activity was …
